HISTOLOGY OF SHOOT BUD FORMATION IN CALLUS CULTURE OF AN OIL YIELDING PLANT: NIGER (GUIZOTIA ABYSSIN
When callus from the hypocotyl was transplanted to MSO (Murashige and Skoogs medium 1962 without any phytohormone) with various combinations of phytohormones, shoot differentiation in G. abyssinica was found. Serial microtome sections of callus grown on MSO with cytokinins (BAP, KN, and zeatin) demonstrated that callus that seemed unorganised on the outside contained intrinsic cellular differentiation. The commencement of differentiation of the trachiedal element to shoot bud production occurs within one to three weeks of callus subculture, according to sections of callus of various ages. The production of shoot buds was clearly exogenous.
