EMBRYOGENESIS AND PLANT REGENERATION FROM TISSUE CULTURE OF SEHIMA NERVOSUM (ROTTL.) STAPF | BIONATU

From mature caryopses of Sehima nervosum cultivated on Murashige and Skoog's (1962) media supplemented with 3.0 mgl-1 2,4-dichlorophenoxyacetic acid, a white, compact, hard, nodular embryogenic callus was formed. MS medium containing 2.0 mgl-1 2,4-D and 100 mgl-1 ascorbic acid was used to maintain these embryogenic cells. When transferred to MS media with 1.0 mg-1 2,4-D under dark conditions, embryogenic cultures organised into nicely defined grass embryos. These embryos matured into whole plantiets after being moved to auxin-free MS media and preserved in the light. 980 (Ca 98 percent) of 1000 embryos transplanted for germination developed full plantlets. In the greenhouse, young plantlets were successfully transplanted to pots and nurtured to maturity. Except for ten albino plants, all of the plants were green.
