CLONAL EVOLUTION, IMMUNE RESPONSE AND CANCER | BIONATURE

Observations of sunlight-related events in the skin of over 2000 people, mostly of European origin, were made in Sydney, Australia (34° S. Lat.) beginning in 1964. Many new associations were discovered after counting pigmented lesions. To explain these associations, a theory of spontaneous or UVL-induced somatic mutation was proposed, with evidence of immune rejection responses to benign or malignant tumours detected. A theory of tumour formation initiated by somatic mutation of normal tissue differentiation genes was proposed: cells barred from final differentiation may fail to recognise normal homoeostatic growth controls in many circumstances. It was also hypothesised that such differentiation genes exist in all cell types and that various phacomatosis tumours are classified based on their link to such genes. This paper examines several facets of the author's original idea and highlights areas of that theory that have been mostly overlooked in the last 25 years or so. In his opinion, some present dogmas are deceptive when applied in detail.
