CALLUS FORMATION FROM PROTOPLAST CULTURES OF ALLIUM WAKEGI | BIONATURE

Protoplasts were extracted enzymatically from leaves taken from amphidiploid A. wakegi bulb cultures, and protoplasts were inoculated in KM8P liquid medium with 0.1M Mannitol and 0.4m glucose. Incubated in the dark with 0.4 mg/12,4-D, 2 mg/1NAA, and 1 mg/1BAP. Within two weeks of inoculation, colonies formed, and after one month, they transformed into mini-calluses. The minicallus was grown into a friable callus in MS medium containing 3 percent sucrose, 0.4 mg/1 2,4-D, 2 mg/1 NAA, 1 mg/1 BAP, and 0.8 percent agarose.
