AN EFFICIENT IN VITRO MULTIPLE SHOOTING METHOD OF STEVIA REBAUDIANA BERTONI. | BIONATURE

The existence of stevioside (diterpene glycoside), a strong alternative to sucrose, makes the little miracle plant Stevia rebaudiana Bert. commercially valuable. Only vegetative propagation by stem cuttings is possible due to poor seed germinability. As a result, micropopulation is the most reliable strategy. Cotyledonary leaves grown axenically yield numerous calli (10mg/lit NAA-0.5mg/lit Kinetin), which develop into microshoots. At moderate concentrations of Kinetin and BA (Kinetin 3mg/lit-5mg/lit, BA 0.5mg/lit-3mg/lit), vigorous microshooting (max 72.65, microshoots/explant) was observed, with maximal shoot length in the shortest shoot initiation time. At high IBA concentrations (5mg/lit- 10mg/lit), most of these microshoots rooted efficiently (20 roots per shoot; 4cm root length) within 5-6 days of incubation. Routine hardening techniques are used to assess the acclimatisation and field establishment capability of such efficient microshoots. The study's goal is to create an elite line (microshoots) with good regeneration, field transfer capability, and long-term stevioside production.
